چکیده
Copine3, a known calcium-dependent membrane binding protein, contains two tandem C2 domains and an A domain. This protein has been shown to interact with receptor tyrosine kinase 2 (ErbB2), but little is known concerning the physiological function of Copine3. To better understand its cellular function, we carried out a yeast twohybrid screen to find Copine3 binding partners. Among the identified proteins, Jun activation domain-binding protein 1 (Jab1) appears to directly interact with Copine3. This physical interaction between Copine3 and Jab1 as well as the specific binding regions of both proteins were confirmed in vitro and in vivo. Our results also demonstrate that binding of Copine3 to ErbB2 is increased when Jab1 is overexpressed in SKBr3 breast cancer cells. Furthermore, two ErbB2 downstream signaling proteins [phosphatidylinositol 3 (PI3) kinase and protein kinase B (AKT)] were also activated by Jab1 overexpression in these cells. These data suggest that binding of Copine3 and Jab1 regulates, at least to some extent, the ErbB2 signaling pathway. Moreover, overexpression of both Copine3 and Jab1 in SKBr3 cells effectively increased cellular migration. Collectively, our findings indicating that Jab1 enhances the ErbB2 binding ability of Copine3, further activating the ErbB2 signaling pathways involved in breast cancer cell pathogenesis.

Background: Staphylococcus aureus and Staphylococcus epidermidis are major human pathogens of increasing importance due to the spread of antibiotic resistance. Novel potential targets for therapeutic antibodies are products of staphylococcal genes expressed during human infection. Previously, the secreted and surface-exposed proteins among seroreactive antigens have been discovered. Furthermore...
متن کاملIn Silico Characterization of Proteins Containing ARID-PHD Domain and Its Expression in Aeluropus littoralis Halophyte
Abiotic stresses are the most important factors that reduce the yield of crops. In this case, Bioinformatics analysis plays an important role to study genes, and their relatedness as well as prediction their function in response to abiotic stresses. Among all domains, ARID-PHD domain has been identified in plants and animals and has a very significant role in growth regulation, cell cycle, and ...
متن کاملCopines-1, -2, -3, -6 and -7 show different calcium-dependent intracellular membrane translocation and targeting.
The copines are a family of C2- and von Willebrand factor A-domain-containing proteins that have been proposed to respond to increases in intracellular calcium by translocating to the plasma membrane. The copines have been reported to interact with a range of cell signalling and cytoskeletal proteins, which may therefore be targeted to the membrane following increases in cellular calcium. Howev...
متن کاملProtein phosphatase 2B (PP2B, calcineurin) in Paramecium: partial characterization reveals that two members of the unusually large catalytic subunit family have distinct roles in calcium-dependent processes.
We characterized the calcineurin (CaN) gene family, including the subunits CaNA and CaNB, based upon sequence information obtained from the Paramecium genome project. Paramecium tetraurelia has seven subfamilies of the catalytic CaNA subunit and one subfamily of the regulatory CaNB subunit, with each subfamily having two members of considerable identity on the amino acid level (>or=55% between ...
متن کاملArabidopsis leaf plasma membrane proteome using a gel free method: Focus on receptor–like kinases
The hydrophobic proteins of plant plasma membrane still remain largely unknown. For example in the Arabidopsis genome, receptor-like kinases (RLKs) are plasma membrane proteins, functioning as the primary receptors in the signaling of stress conditions, hormones and the presence of pathogens form a diverse family of over 610 genes. A limited number of these proteins have appeard in pr...
